Speak to an equine insurer, they offer standard policies for towing.
Hiring your trailer out for money would not be covered under a normal policy as it consitutes a commercial arrangement. Also taking your friends horses somewhere in return for money is a commercial arrangement and requires an operators licence.

Your friend needs to be insured.. under her own insurance policy.. and do be careful.. and make it all clear before you do this.. I have had it go wrong..

I've just got a trailer, so for yourself you'll need to check in 3 areas:
Your trailer - that will need insuring for fire/theft/3rd party etc, all the usuall stuff Your car/towing vehicle - check that your insurance for that covers you towing a trailer, some policies don't, it's a way of making them cheaper. Your horse - check that your horse insurance covers travelling in a trailer!
If you take your friend as Susan has found to her cost make sure that she is insured so that if her horse is injured or damages your trailer you are not out of pocket. Friendly agreements can go badly wrong if a lot of money is involved.
If you hire out then that is a whole different ball game. Even if you just hire out the trailer, all your normal insurance will not cover you for hire for reward. You will need specialist stuff. If you provide the whole transport service (ie: not just a trailer for them to tow) you will need an operators licence. This all adds up to a lot of extra expense which for most people just will not be worth it.
